From 1938 to 1944 Castellanos studied at the Superior School of Music of Caracas. In 1947 he went to New York City on a two-year scholarship studying piano with Carlos Buhler. He held various teaching positions in Caracas and founded the Collegium Musicum de Caracas, a Venezuelan musical group. Alba Quintanilla was among his pupils.
